That really depends on a lot of factors. It won't disconnect a TCP
connection that's currently active. However, you run a risk that a UDP
conversation might get disrupted, depending on the timing.
And, if domain lookups are essential for establishing outgoing
connections (which they would be, unless the connections are all to
hosts in the hosts table, or unless they're made by IP address) then you
may disable the ability to create new connections while you're making
the change.
Worse, if there's a problem with the new configuration. (I.e. the new
DNS server doesn't work well, or is missing some necessary local hosts,
etc) you could break the ability to establish new connections until the
error is discovered and fixed.
With what's at stake, I'd consider testing it during off hours, or at
least on a test box before changing the production box.
The most dangerous bugs are always the ones you don't think of.
